A police spokesperson said police were called to the truck, alight, about 9.30pm.
The prime mover was fully loaded with cattle about 40km out the South Condobolin road when a tyre blew and the fire was sparked, the spokesperson said.
The trailer caught fire and two of the cattle were killed. The rest of the cattle were saved.
Firefighters attended and extinguished the fire.
